Bio-based and cost effective method for phenolic compounds removal using cross-linked enzyme aggregates

This work aimed at presenting a green method using new source of peroxidase isolated from Raphanus sativus var. niger (RSVNP) in immobilized form, for the treatment wastewater. To ensure stability and enzymatic activity biodegradation process, RSVNP was as cross-linked enzyme aggregate (CLEAs). With more than 29% recovered 85% aggregation yield, acetone selected best precipitating agent. The formed protein aggregates required 2% (v/v) glutaraldehyde (GA) concentration ratio 9:1 (E) amount to cross-linker (E/GA). Compared free enzyme, RSVNP-CLEAs were found chemically thermally stable exhibited good storage 8 weeks. In addition, evaluated their ability remove phenol p-cresol aqueous solution by varying several operating conditions. A maximal yield (98%) conversion recorded after 40 min; while 92% degraded 1 h duration time. reusability tested, displaying 71% degradation third batch carried out 54% achieved four successive reuses presence hydrogen peroxide 2 mM concentration.
